In a game with more than one jester (i.e. a 4-player game), a jester in the mission zone does NOT prevent other jesters from being played and applying their rule.
The jester is not a class ability but rather a special rule that triggers when a Jester is played. A jester in the mission zone has no effect - lucky you!
